What is the definition of a n.i.g.g.e.r and why is it considerred an offensive word?

I have sen Afro-American dark skinnned perfectly fine individuals liberally use this term to define themselves but they take huge offense if someone else calls them by this name. Could you please tell me how it originated and why is this word considered offensibe if an American calls them by this name?
